Description:
The ALF-161 antibody reacts with mouse interleukin-1 alpha (IL-1α), also known as Lymphocyte Activating Factor (LAF), Endogenous Pyrogen (EP), Leukocyte Endogenous Mediator (LEM), and Mononuclear Cell Factor (MCF). IL-1α is a 17 kDa factor produced by macrophages, dendritic cells, and lymphocytes. It primarily exists in a cell-associated form and shares 23% amino acid homology with IL-1β. IL-1α regulates immune responses in various cell types and induces fever, hypotension, and acute-phase reactions in vivo.

Applications Reported:
The ALF-161 antibody is suitable for ELISA capture, ELISPOT capture, neutralization, and intracellular staining for flow cytometry.

Applications Tested:
The Functional Grade Purified ALF-161 antibody has been validated by LAL assay (low endotoxin), ELISA, and bioassay for IL-1α neutralization.

• At 0.031 µg/mL, the antibody inhibits 50% of the biological activity of 4.0 ng/mL mouse IL-1α in a D10 cell proliferation assay.
• For ELISA capture, a suitable range is 1–4 µg/mL using recombinant mouse IL-1α as the standard (1000 pg/mL – 8 pg/mL range).

Target Information

IL-1 alpha (Interleukin-1 alpha):
A proinflammatory cytokine expressed by monocytes, macrophages, and dendritic cells. Encoded by the IL1A gene on chromosome 2q13, IL-1α signals through IL-1RI and IL-1RII receptors shared with IL-1β. It plays a central role in innate immunity by inducing other cytokines and acute-phase responses. IL-1α activity is regulated by IL-1 Receptor Antagonist (IL-1RA), which blocks receptor binding. Elevated IL-1α levels are associated with chronic inflammatory diseases such as rheumatoid arthritis, psoriasis, and multiple sclerosis.
