Target Information
GLTSCR2 (also known as NOP53) is a nucleolar protein involved in integrating the 5S RNP into the ribosomal large subunit during ribosome biogenesis. It may also participate in rRNA transcription and act as a nucleolar sensor regulating p53/TP53 activation in response to ribosome biogenesis perturbation, DNA damage, and other stress conditions.
Disruption of the NOP53–RPL11 interaction allows RPL11 transport to the nucleoplasm, inhibiting MDM2 and activating p53/TP53. GLTSCR2 may also enhance p53-mediated cell cycle arrest and apoptosis by preventing MDM2-dependent degradation. Identified as a tumor suppressor, it stabilizes PTEN, antagonizing the PI3K-AKT signaling pathway, and may inhibit cell proliferation through interaction with NF2. Additionally, it regulates NPM1 localization and degradation, thereby modulating MYC–NPM1-mediated transcription. GLTSCR2 may also influence cellular aerobic respiration and attenuate interferon-beta response via DDX58/RIG-1 inhibition.
