Product Specific Information

The 61D3 monoclonal antibody reacts with human CD14, a 53–55 kDa GPI-linked glycoprotein expressed on monocytes, interfollicular macrophages, and some dendritic cells. CD14 binds LPS-LBP complexes with high affinity on monocytes.

  • Applications Reported: Flow cytometric analysis
  • Applications Tested: Pre-diluted and validated for flow cytometry using normal human peripheral blood cells
  • Recommended Use: 5 µL (1.0 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test)

Brilliant Violet™ 786 (BV786) emits at 786 nm and is optimized for cytometers equipped with a violet (405 nm) laser. Ensure instrument compatibility.

When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401-42) or Brilliant Stain Buffer™ (Product # 00-4409-75) to minimize non-specific interactions.

Light Sensitivity: Protect vial and stained samples from light.
Fixation: Compatible with IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54) for up to 3 days at 4°C.
Not compatible with methanol-based fixation.

Target Information

CD14 is a 55 kDa GPI-anchored glycoprotein expressed on mature monocytes, macrophages, and neutrophils. It functions as a lipopolysaccharide receptor and is released into serum as soluble sCD14. CD14 plays roles in immune response, apoptosis recognition, and inflammatory regulation. Alternative splicing produces multiple transcript variants of the same isoform.
Diseases associated with CD14 dysfunction include Mycobacterium chelonae infection and Croup.
