Product Specific Information

Description:
The IP26 monoclonal antibody reacts with the human alpha beta TCR dimer, which is expressed by the majority of peripheral T cells.

Applications Reported:
This IP26 antibody has been reported for use in flow cytometric analysis.

Applications Tested:
Pre-diluted and tested by flow cytometric analysis of normal human peripheral blood cells. Use at 5 µL (0.5 µg) per test.
Each test is defined as the amount of antibody that will stain a cell sample in a final volume of 100 µL.
Cell number can range from 10⁵ to 10⁸ cells/test.

Fluorochrome Information:
Super Bright 436 can be excited with the violet laser line (405 nm) and emits at 436 nm.
A 450/50 bandpass filter (or equivalent) is recommended.
Ensure instrument compatibility for this fluorochrome.

Staining Buffer Recommendation:
When using two or more Super Bright dye–conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

405 nm / 436 nm / Violet Laser

Target Information

The T cell antigen receptor (TCR) consists of a ligand-specific alpha/beta heterodimer associated with invariant CD3 subunits, essential for surface expression.
TCR engagement induces T cell activation, differentiation, and cytokine production.
Most T cells in blood and lymphoid organs express alpha/beta TCRs, while gamma/delta TCRs are mainly found in epithelial tissues.
The TCR-CD3 complex formation requires all subunits for proper expression.
TCR recognition of self-peptides is linked to autoimmune diseases, while mutant peptides are associated with tumors.
Superantigen engagement of TCRs can cause toxic shock syndrome.
Autoantibodies to TCR V beta segments have been observed in RA and SLE, potentially modulating immune responses.
Gamma-delta T cells show limited diversity and distinct antigen recognition, with subsets exhibiting antitumor and immunoregulatory activity.
