Product Specific Information
The J43 monoclonal antibody reacts with mouse PD-1 (programmed death-1), a 55 kDa member of the Ig superfamily. PD-1 contains the immunoreceptor tyrosine-based inhibitory motif (ITIM) and plays a key role in peripheral tolerance and autoimmune disease in mice. PD-1 is expressed mainly on activated T and B lymphocytes.
Two novel B7 family members, PD-L1 (B7-H1) and PD-L2 (B7-DC), serve as PD-1 ligands. J43 inhibits the binding of mouse PD-L1-Ig and PD-L2-Ig to PD-1/BHK transfected cells. In vivo, J43 enhances contact hypersensitivity and exacerbates acute GVHD, EAE, and NOD diabetes, similar to PD-1-deficient models.

Applications Tested: Tested on stimulated mouse splenocytes (≤1.0 µg/test).
A test is defined as the amount of antibody staining a cell sample in 100 µL volume. Optimal titration is recommended.
Fluorochrome Information:
Super Bright 436 is excited with the violet laser (405 nm) and emits at 436 nm. Use a 450/50 bandpass filter or equivalent.
For panels using multiple Super Bright dyes, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific interactions.

Target Information
T-cell activation is self-limited by inhibitory receptors such as PD-1 (PDCD-1), which binds ligands PD-L1 and PD-L2. Upon ligand binding, PD-1 transmits inhibitory signals that modulate immune responses. PD-1 deficiency leads to hyperactivated immune systems and autoimmune diseases. Despite its predicted molecular weight, PD-1 often migrates at higher molecular weight in SDS-PAGE.
