Target Information
Adeno-Associated Virus capsid protein self-assembles to form an icosahedral capsid with T=1 symmetry, approximately 22 nm in diameter. The capsid consists of 60 copies of three variants—VP1, VP2, and VP3—which differ in their N-terminus. It encapsulates genomic ssDNA and binds to host cell heparan sulfate, using host ITGA5-ITGB1 as a coreceptor for virion attachment. This attachment triggers internalization via clathrin-dependent endocytosis. Receptor binding induces capsid rearrangement, exposing the VP1 N-terminus, including its phospholipase A2-like region and nuclear localization signal(s). The VP1 N-terminus may act as a lipolytic enzyme to breach the endosomal membrane and assist in nuclear transport of the virus.
