Target Information

Phosphofructokinase (PFK) is a major regulatory enzyme involved in cellular sugar metabolism, crucial for energy production and normal cell function. In humans, three structural loci—M (muscle), L (liver), and P (platelet)—control PFK subunits, expressed variably across tissues. Human fibroblasts and leukocytes express all three genes. PFK forms tetramers through random association of subunit products, resulting in five isozyme forms. Muscle and liver PFKs are homotetramers (M4, L4), while red cells contain all five isozymes (M4, M3L, M2L2, ML3, L4).

PFK acts as an allosteric enzyme activated by ADP, AMP, or fructose bisphosphate and inhibited by ATP or citrate. It catalyzes the rate-limiting step in glycolysis. PFK deficiency can cause mild to severe episodic illness, often presenting with dark-colored urine after exercise. Elevated red cell PFK levels are observed in individuals with Down syndrome.
