Product Specific Information

For reconstitution, add 100 µL of distilled water to achieve a final antibody concentration of approximately 1 mg/mL.
For conjugation experiments, it is recommended to perform an additional desalting step (e.g., Zeba Spin Desalting Columns, 7K MWCO, 0.5 mL, Product #89882).

Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nucleotide deprivation. p53 upregulation leads to activation of genes such as p21waf1, resulting in G1 or G2/M arrest.

p53 structure includes:

  • N-terminal transactivation domain
  • Central DNA-binding domain
  • Oligomerization domain
  • C-terminal regulatory domain

Phosphorylation at Ser15 is crucial for p53 activation and stabilization. p53 plays a key role in preventing proliferation of damaged cells and acts as an anticancer factor. Phosphorylation at Ser392 is associated with human tumor formation. p53 is also linked to aging, oxidative stress, and cognitive function (LTP regulation).

While p53 is expressed at low levels in normal cells, it is highly expressed in transformed cell lines, contributing to malignancy. Mutations in the TP53 gene disrupt DNA binding and tumor suppressor function, and are found in many human cancers as well as in germline mutations (e.g., Li-Fraumeni syndrome).
