Specificity Comments:
This antibody recognizes a cell surface glycoprotein of 95/115/135 kDa (depending on glycosylation), identified as CD43. CD43 is expressed in 70–90% of T-cell lymphomas and 22–37% of B-cell lymphomas, but not in reactive B-cells. Therefore, co-expression of CD43 in a B-lineage population suggests malignant lymphoma rather than a reactive process. When used with anti-CD20, CD43 enables effective immunophenotyping of lymphomas in formalin-fixed tissues. Co-staining with anti-CD20 and anti-CD43 supports a lymphoma diagnosis over a reactive lymphoid process.

Target Information

CD43 (leukosialin, sialophorin) is a transmembrane mucin-like protein with a high negative charge, expressed on most hematopoietic cells. It contributes to a repulsive barrier that interferes with cellular adhesion and can promote leukocyte aggregation. Through interactions with actin-binding proteins ezrin and moesin, CD43 regulates T-cell morphology and cell-cell interactions during lymphocyte trafficking. CD43 signaling enhances LFA-1 adhesiveness and modulates immune cell activity. It can induce tumor suppressor p53 expression, but in the absence of p53 and ARF, promotes tumor proliferation. CD43 plays an important role in leukocyte function and is implicated in disorders such as Wiscott-Aldrich Syndrome and Adenoid Basal Cell Carcinoma.
