Immunohistochemistry (PFA fixed): incubate antibody for 30 minutes at room temperature.
Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

CD43 (leukosialin, sialophorin) is a transmembrane mucin-like protein with a high negative charge, expressed on the surface of most hematopoietic cells.
It acts as a repulsive barrier interfering with cellular adhesion and can promote leukocyte aggregation.
By interacting with actin-binding proteins ezrin and moesin, CD43 regulates T-cell morphology and cell-cell interactions during lymphocyte traffic.
CD43 signaling enhances LFA-1 adhesiveness while counteracting its induction via other receptors.
Expression of CD43 induces functionally active tumor suppressor p53 protein, but in p53 and ARF deficiency, CD43 promotes tumor proliferation and viability.
CD43 is an important modulator of leukocyte functions.
Diseases associated with CD43 dysfunction include Wiscott-Aldrich Syndrome and Adenoid Basal Cell Carcinoma.
