Target Information
Tumor necrosis factor (TNF) receptor-associated factors (TRAFs) are adaptor proteins linking the TNF receptor superfamily to signaling pathways, regulating cell death and stress responses. TRAF proteins share a homology region enabling receptor and TRAF–TRAF interactions, activating various signal cascades. TRAF2 and TRAF3 bind CD40, crucial for NF-κB activation in mouse B lymphocytes. TRAF2 and TRAF6 are required for CD40 signaling in nonhematopoietic cells. TRAF2 also interacts with lymphotoxin-beta receptor (LTβR) with TRAF3 and apoptosis inhibitors cIAP1 and Smac.
