Target Information

TLR4 (Toll-like receptor 4) is a member of the Toll-like receptor family, playing a key role in pathogen recognition and activation of innate immunity. These receptors are highly conserved across species and are structurally and functionally similar. TLR4 is expressed abundantly in the placenta and in the myelomonocytic subpopulation of leukocytes. It mediates cellular responses to lipopolysaccharide (LPS) by forming a receptor complex with CD14 and LY96, initiating signaling pathways that lead to cytokine production and immune activation. Mutations in TLR4 can alter LPS responsiveness, and several transcript variants exist, though not all have confirmed protein-coding potential. TLR4-deficient mice demonstrate increased susceptibility to Gram-negative bacterial sepsis.
