Product Specific Information

Heat-mediated antigen retrieval is recommended prior to staining, using a 10 mM citrate buffer (pH 6.0) for 10 minutes followed by cooling at room temperature for 20 minutes. After antigen retrieval, incubate samples with primary antibody for 30 minutes at room temperature. A suggested positive control is tonsil tissue.


Target Information

CD43 (leukosialin, sialophorin) is a transmembrane mucin-like protein with a high negative charge, expressed on the surface of most hematopoietic cells. It contributes to a repulsive barrier interfering with cellular adhesion but can also promote leukocyte aggregation. Through interactions with actin-binding proteins ezrin and moesin, CD43 regulates T-cell morphology and cell-cell interactions during lymphocyte traffic. CD43 signaling enhances LFA-1 adhesiveness and counteracts its induction via other receptors. Expression of CD43 induces functionally active tumor suppressor p53 protein; however, in the absence of p53 and ARF, CD43 promotes tumor proliferation and viability. CD43 is an important modulator of leukocyte functions. Diseases associated with CD43 dysfunction include Wiscott-Aldrich Syndrome and Adenoid Basal Cell Carcinoma.
