Target Information
Glutathione peroxidases (GPXs) are ubiquitously expressed enzymes that catalyze the reduction of hydrogen and organic hydroperoxides by glutathione. Multiple isoforms exist, differing in structure and localization:
- GPX1 (cGPx): Cytosolic/mitochondrial, selenium-dependent enzyme, first discovered member of GPX family.
- GPX2 (GI-GPx): Expressed in gastrointestinal epithelium.
- GPX3 (pGPx): Extracellular plasma form, mainly expressed by the kidney and released into blood circulation.
- GPX4 (PH-GPx): Reduces various hydroperoxides, including those in membranes and lipoproteins.
All mammalian GPX family members, except for Cys-containing GPX3 and epididymis-specific GPX5 isoforms, possess selenocysteine at the active site.
