Target Information

ALAS-1 is a nuclear-encoded mitochondrial matrix enzyme that catalyzes the condensation of glycine with succinyl-CoA to form delta-amino levulinate, CO₂, and CoA. It regulates the first and rate-limiting step of the heme biosynthetic pathway.
ALAS1 is a pyridoxal-5'-phosphate-dependent housekeeping enzyme, ubiquitously expressed, and provides heme for cytochromes and other hemoproteins.
In the liver, ALAS1 undergoes negative feedback regulation by heme. Its transcription is stimulated by cAMP and respiratory uncoupling, while phorbol esters and insulin repress expression.
ALAS1 is implicated in the reciprocal regulation of heme biosynthesis and the circadian clock, suggesting potential therapeutic relevance for circadian disorders. Downregulation of ALAS1 expression in acute liver failure is associated with altered heme metabolism and impaired liver function.
