Product Specific Information

The UCHT1 monoclonal antibody reacts with human CD3e, a 20 kDa subunit of the TCR complex. Along with other CD3 subunits (gamma, delta), the epsilon chain is required for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes and all mature T cells.

Crosslinking of TCR via immobilized UCHT1 initiates intracellular signaling leading to cellular activation and proliferation.

Each product includes:

• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells (5 µL (0.4 µg)/test). A test is defined as the amount of antibody that stains a cell sample in 100 µL volume (10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
• Add 5 µL per sample for antibody cocktails or single-color controls.

NovaFluor conjugates utilize Phiton technology with engineered fluorescent signatures optimized for minimal spectral spillover.

Target Information

The CD3 subunit complex transduces antigen-recognition signals into T-cell cytoplasm and regulates TCR surface expression. It includes CD3 gamma, delta, epsilon, and zeta subunits, encoded on human chromosome 11. These subunits associate with cytoplasmic signaling molecules through tyrosine-based motifs.

CD3 is expressed on:

• 68–82% of normal peripheral blood lymphocytes
• 65–85% of thymocytes
• Purkinje cells in the cerebellum

Not expressed on B or NK cells. CD3 gene defects are linked to CD3 immunodeficiency.
