Target Information
MYLK (MLCK) is a serine/threonine kinase that phosphorylates myosin regulatory light chains to facilitate myosin–actin interaction and contractile activity. It plays a key role in smooth muscle contraction, vascular permeability, and tissue edema formation.
In the nervous system, MLCK regulates astrocytic process growth and neurotransmitter release at sympathetic synapses. It also participates in fibroblast apoptosis signaling.
Smooth muscle and non-muscle isoforms are widely expressed in adult and fetal tissues, with non-muscle isoform 2 being predominant. The Telokin isoform binds calmodulin and is also broadly expressed. MLCK activity is likely down-regulated by phosphorylation and contains one fibronectin type III domain and nine immunoglobulin-like C2-type domains.
