Product Specific Information

Description:
The 145-2C11 monoclonal antibody reacts with mouse CD3e, a 20 kDa subunit of the TCR complex. The epsilon chain, along with gamma and delta subunits, is essential for proper TCR assembly, trafficking, and surface expression. CD3 is expressed by thymocytes and all mature T cells. Binding of 145-2C11 to TCR initiates intracellular activation, proliferation, or apoptosis, depending on assay conditions. It is widely used as a phenotypic marker for mouse T cells.

Each product includes:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes at ≤1.0 µg/test (100 µL final volume). Optimal titration recommended.

Important Notes:

  • NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
  • CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal staining.
    • Add 5 µL per sample before mixing with cells.
    • For single-color controls, use 5 µL per 100 µL sample (10³–10⁸ cells).

Target Information

The CD3 subunit complex is essential for transducing antigen-recognition signals into T cells and regulating TCR surface expression. It comprises CD3 gamma, delta, epsilon, and zeta subunits, all members of the immunoglobulin superfamily encoded on chromosome 11. These subunits associate with signal transduction molecules via tyrosine-based motifs. CD3 plays roles in TCR-induced growth arrest, survival, and proliferation.

  • Expression: 68–82% of peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells in the cerebellum.
  • Not expressed on: B or NK cells.
  • Clinical relevance: Decreased T lymphocytes may occur in autoimmune diseases.
  • Genetic location: CD3 epsilon, gamma, and delta genes on chromosome 11.
  • Defects: Associated with CD3 immunodeficiency.
