Product Specific Information

The 145-2C11 monoclonal antibody reacts with mouse CD3e, a 20 kDa subunit of the TCR complex. CD3e, along with gamma and delta subunits, is essential for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes and mature T cells. Binding of 145-2C11 to TCR can trigger cellular activation, proliferation, or apoptosis depending on experimental conditions. Commonly used as a phenotypic marker for mouse T cells.

Each product contains:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Applications Tested: Tested on mouse splenocytes (≤1.0 µg/test). Optimize antibody titration for best performance.

Note:
NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.

This conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), which improves staining performance. Add 5 µL per sample to antibody cocktails before combining with cells.

NovaFluor conjugates utilize Phiton™ technology, enabling engineered fluorescent signatures with reduced spillover and spread.

Target Information

The CD3 subunit complex is crucial in transducing antigen-recognition signals into the cytoplasm of T cells and regulating TCR surface expression. Activation involves CD3 gamma, delta, epsilon, and zeta subunits, which contain cytoplasmic tails with signaling motifs. CD3 is expressed on most T lymphocytes and thymocytes but not on B or NK cells. Defects in CD3 genes are associated with immunodeficiency.
