MLH1 is a DNA mismatch repair protein essential for maintaining genetic integrity. Defects in mismatch repair pathways lead to microsatellite instability (MSI), which is associated with carcinogenesis.
Mutations in mismatch repair genes, including MLH1, are linked to hereditary nonpolyposis colon cancer (HNPCC). MSH2 binds to mismatched DNA duplexes and recruits MLH1 and PMSH heterodimers, facilitating subsequent repair steps.
