Product Specific Information
The monoclonal antibody 29A1.4 recognizes mouse NKp46 (CD335), a member of the natural cytotoxicity receptor (NCR) family. CD335 is a glycoprotein with two Ig-like domains and a short cytoplasmic tail, uniquely expressed on NK cells (including immature NK cells, DX5⁻ CD3⁻), allowing discrimination between NKT and NK cells (NKp46⁺, CD3⁻).
Unlike many NK markers that also stain NKT cells, staining with 29A1.4 is not strain-specific and has been demonstrated in C57Bl/6, SJL, CBA/CA, and BALB/C strains. NKp46 plays a role in NK cell-mediated lysis of tumor and pathogen-infected cells.
- The 29A1.4 antibody activates NK cells in vitro but does not deplete NK cells in vivo.
- Reported application: Flow cytometric analysis.
- Tested on mouse splenocytes; use ≤1 µg/test (100 µL final volume).
- Cell number range: 10⁵–10⁸ cells/test.
- Optimize antibody titration for best performance.
Light Sensitivity: PE-Cyanine7 is sensitive to photo-induced oxidation. Protect from light.
Fixation: Samples can be stored up to 3 days at 4°C in the dark using:
- IC Fixation Buffer (Product #00-8222)
- 1-step Fix/Lyse Solution (Product #00-5333)
Excitation: 488–561 nm
Emission: 775 nm

Target Information
NKp46 (CD335, NCR1) is a cytotoxicity-activating receptor that enhances the ability of activated NK cells to mediate tumor cell lysis. It consists of two Ig-like domains and is structurally related to leukocyte immunoglobulin-like (LIR) and killer inhibitory receptors (KIR).
