Product Specific Information

The eBioGoH3 monoclonal antibody reacts with mouse and human CD49f (Integrin alpha 6), also known as VLA-6 alpha chain and platelet gpIc. CD49f is a 120 kD transmembrane protein that associates with CD29 (Integrin β1) to form VLA-6, and with CD104 (Integrin β4) to form α6β4 complex. CD49f is expressed on T cells, monocytes, platelets, epithelial, and endothelial cells, as well as germinal center B cells. This antibody cross-reacts with human, mouse, and bovine integrin α6 and can block its binding to laminin.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-diluted and validated for flow cytometry using normal human peripheral blood cells at 5 µL (0.06 µg)/test. One test corresponds to staining in a 100 µL final volume, with 10⁵–10⁸ cells/test.

Fluorophore Information:
Super Bright 600 is a tandem dye excited by the 405 nm violet laser and emits at 600 nm. Recommended detection using a 610/20 bandpass filter. Ensure instrument compatibility.

Staining Recommendations:
For panels using multiple Super Bright dyes, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Light Sensitivity:
Protect vial and stained samples from light due to photo-induced oxidation sensitivity.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark with minimal loss of brightness or FRET efficiency.

Target Information

CD49f (Integrin alpha-6, ITGA6) is an integrin alpha chain protein forming heterodimers with various beta chains (e.g., β1 to form VLA-6 or β4 to form TSP180). Integrins mediate cell adhesion and cell-surface signaling, linking the extracellular matrix (ECM) to intracellular cytoskeletal proteins. They regulate cell shape, motility, proliferation, and survival. CD49f is mainly expressed in epithelial cells and plays a structural role in hemidesmosomes. Over 18 alpha and 8 beta integrin subunits with multiple isoforms exist in mammals, some binding soluble ligands or counter-receptors like ICAMs, facilitating cell aggregation.
