Product Specific Information

The HM48-1 monoclonal antibody reacts with the mouse CD48 antigen (also known as BCM1, Blast-1, OX-45).
CD48 is a 45 kDa GPI-linked glycoprotein expressed on most hematopoietic cells and is part of the SLAM and Ig superfamily.
It is useful for distinguishing hematopoietic progenitor subsets:

• HSC: CD150(+)CD244(-)CD48(-)
• MPP: CD244(+)CD150(-)CD48(-)
• Restricted progenitors: CD48(+)CD244(+)CD150(-)

CD48 plays key roles in adhesion and T cell activation. In mice, CD2 and CD244 act as counter-receptors.
HM48-1 modulates CD48 functions by blocking CD48/CD2 and CD48/CD244 interactions, inhibiting mitogen-induced proliferation, providing costimulatory signals, and prolonging cardiac allograft survival.

Applications Tested: Mouse splenocytes, ≤ 0.5 µg/test (100 µL final volume, 10⁵–10⁸ cells/test)

Fluorochrome Information:
Super Bright 600 is a tandem dye excited by the 405 nm violet laser and emits at 600 nm. Recommended filter: 610/20 bandpass.
Use Super Bright Complete Staining Buffer (Product # SB-4401) when combining multiple Super Bright dyes to minimize non-specific interactions.
Protect from light due to photo-oxidation sensitivity.

Fixation:
Samples can be stored up to 3 days at 2–8°C in the dark using:

• IC Fixation Buffer (Product # 00-8222)
• 1-step Fix/Lyse Solution (Product # 00-5333)

Excitation: 405 nm
Emission: 600 nm

Target Information

CD48 (Blast-1) is a member of the CD2 subset of the Ig superfamily, including CD2, CD58, CD84, CD150, CD229, and CD244.
It mediates homotypic and heterotypic adhesion and acts as a high-affinity ligand for 2B4 and low-affinity ligand for CD2.
CD48 is expressed on lymphocytes and monocytes, weakly on granulocytes, and absent on platelets, fibroblasts, and endothelium.
It is a marker for GPI-anchor defects (e.g., PNH) and may play roles in allergic airway diseases such as rhinitis and asthma.
