Target Information
CD163 (also known as M130 antigen, Ber-Mac3, Ki-M8, SM4) is a 130 kDa membrane glycoprotein belonging to the scavenger receptor cysteine-rich superfamily. It functions as a receptor for the hemoglobin-haptoglobin complex and protects tissues from oxidative damage caused by free hemoglobin. CD163 participates in iron recycling through endocytosis and heme breakdown.
This protein is expressed on human monocytes and macrophages, particularly during the late phase of inflammation. It is found on all circulating monocytes and most tissue macrophages, excluding those in the mantle zone and germinal centers of lymphoid follicles, interdigitating reticulum cells, and Langerhans cells. CD163 expression is upregulated by IL-10, IL-6, and dexamethasone, while LPS and PMA induce shedding of CD163 into plasma or supernatant. The receptor binds hemoglobin/haptoglobin complexes in a calcium- and pH-dependent manner, with higher affinity for multimeric haptoglobin (HP1F) complexes. CD163 activation triggers intracellular signaling involving tyrosine kinase-dependent calcium mobilization, inositol triphosphate production, and secretion of IL-6 and CSF1.
