Target Information
FGF2 (fibroblast growth factor basic, FGFb) is a member of the fibroblast growth factor family. It interacts with high-affinity transmembrane receptors to regulate cell proliferation and tissue neovascularization.
FGF2 exists as five isoforms with distinct intracellular localizations and functions:
- 18 kDa isoform: predominantly cytosolic, acts through cell surface receptors
- 22, 22.5, 24, and 34 kDa isoforms: nuclear, may signal independently of receptor pathways
In humans, the gene is located on chromosome 4q. FGF2 is involved in limb and nervous system development, wound healing, and tumor growth.
The mRNA includes multiple polyadenylation sites and can be translated from non-AUG and AUG initiation codons, producing five distinct isoforms.
CUG-initiated isoforms are nuclear and mediate intracrine effects, while AUG-initiated forms are cytosolic and mediate paracrine and autocrine signaling.
FGF2 dysfunction is associated with diseases such as Kaposi Sarcoma and corneal neovascularization.
