Target Information
Caspase 9 (ICE-like apoptotic protease 6, ICE LAP6, apoptotic protease Mch6, apoptotic protease activating factor 3, Apaf3)는 CARD 도메인을 포함하는 peptidase family C14에 속하는 단백질입니다. Caspases는 apoptosis를 매개하는 cysteine protease로, 비활성 전구체 형태로 발현되며 특정 기질을 절단하여 세포사멸을 유도합니다.
Caspase 9는 활성화 시 heterotetramer 형태로 존재하며, 세포질에서 미토콘드리아로 이동합니다. Caspase 9는 apoptosis 실행에 관여하는 caspase 활성화 연쇄반응의 일부로서 Caspase 3와 Caspase 6을 절단 및 활성화합니다. 또한 Apaf-1과 결합하여 caspase-3를 활성화시키며, ABL1/c-Abl 의존적 DNA 손상 유도 apoptosis를 촉진하고 PARP(poly(ADP-ribose) polymerase)를 절단합니다.
Caspase 9는 다양한 인간 조직에서 발현되며, rat의 경우 C-말단이 대체된 isoform이 보고되었습니다. Caspase 9의 변이는 뇌 발달에 영향을 주며, 마우스에서 결핍 시 신경 및 대뇌 발달 이상이 발생합니다. 인간에서는 조직별로 발현이 다르며, 낮은 Caspase 9 수준은 암 및 알츠하이머병과 같은 신경퇴행성 질환과 관련이 있습니다.
