Product Specific Information

The FJK-16s antibody reacts with mouse, rat, dog, porcine, bovine, and cat FOXP3 (FORKHEAD BOX P3, SCURFIN, JM2). FOXP3 is a 49–55 kDa transcription factor, crucial for regulatory T cell (Treg) function. High expression is found in CD4+CD25+ Treg cells, and ectopic expression in CD4+CD25− cells induces a Treg phenotype.

Immunoblotting with FJK-16s maps the epitope to amino acids 75–125 of mouse FOXP3. In humans, this region is alternatively spliced at the mRNA level, leading to isoform diversity not seen in mouse splenocytes.

This antibody is optimized for use with the Foxp3/Transcription Factor Buffer Staining Set (Product #00-5523-00). Use of other buffers is not recommended.

Applications Reported: Intracellular staining followed by flow cytometry.
Applications Tested: Tested by intracellular staining using the Foxp3/Transcription Factor Buffer Set and protocol. Recommended ≤ 0.25 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Optimize titration for best results.

Light Sensitivity: Protect from light due to photo-induced oxidation.
Fixation: Samples can be stored up to 3 days at 4°C in IC Fixation Buffer (#00-8222) or 1-step Fix/Lyse Solution (#00-5333) with minimal fluorescence loss.

Target Information

FOXP3 (Forkhead box protein 3) is a transcriptional regulator essential for immune homeostasis. It is highly expressed in CD25⁺CD4⁺ regulatory T cells, low in CD4⁺CD25⁻ cells, and absent in CD4⁻CD8⁺ cells. FOXP3 mutations cause the scurfy phenotype in mice and IPEX syndrome (immune dysfunction, polyendocrinopathy, enteropathy, X-linked) in humans. FOXP3 expression is also regulated by p53 in cancer cells, influencing immune regulation and tumor progression.
