Target Information

B7-H4 (also known as B7x, VTCN1) is a costimulatory protein that functions as a negative regulator of T-cell mediated immunity. Although its receptor remains unidentified, B7-H4 delivers inhibitory signals to T-cells, preventing proliferation, cell cycle progression, and interleukin-2 production.
B7-H4 deficient mice show minimal immune alteration, suggesting its role in fine-tuning T-cell responses. It is expressed on activated T-cells, B-cells, monocytes, and dendritic cells. Aberrant expression has been linked to lung, breast, and ovarian cancers.
As a membrane-associated immunoinhibitory protein, B7-H4 interacts with T lymphocyte receptors to modulate cellular and humoral immune responses. Overexpression may contribute to tumor immune evasion and progression. VTCN1 is also considered a potential biomarker for early detection of ovarian cancer and renal cell carcinoma (RCC).
