Gene/Protein: TNK2 (ACK) – Activated Cdc42-associated kinase
Function:
TNK2 (ACK1) is a non-receptor tyrosine kinase that signals downstream of Cdc42. It is involved in tumorigenesis and metastasis, particularly in prostate cancer. TNK2 mediates CDC42-dependent cell migration via phosphorylation of BCAR1, and may play roles in synaptic function, plasticity, and brain development. It activates AKT1 by phosphorylation at Tyr-176, and phosphorylates AR, WWOX, and MCF2, promoting cell survival and metastasis. TNK2 also regulates AXL receptor levels and influences tumor growth by modulating tumor suppressors and pro-survival factors.
