TLR4 (Toll-like receptor 4)는 선천 면역 활성화에 중요한 역할을 하는 수용체로, 병원체 인식에 관여합니다. 이 수용체는 사람과 동물 전반에 걸쳐 보존되어 있으며, 주로 태반과 백혈구의 단핵구 아형에서 발현됩니다.
TLR4는 LPS(lipopolysaccharide)를 인식하는 복합체의 일부로, CD14, LY96과 함께 신호 전달을 매개합니다. 이 과정에서 감염원 관련 분자 패턴(PAMPs)을 인식하여 면역 반응에 필요한 사이토카인을 유도합니다.
TLR4 유전자 변이는 LPS 반응성의 차이와 관련이 있으며, 여러 전사 변이체가 존재하지만 대부분의 단백질 발현 가능성은 불확실합니다.
TLR4는 말초 혈액 단핵구 및 일부 B세포에서 발현되며, TLR4 결손 마우스는 LPS 반응성이 결여되어 그람음성균 패혈증에 더 취약함이 보고되었습니다.
