Product Specific Information

The MAb11 antibody reacts with human tumor necrosis factor-alpha (TNF alpha), a 17 kDa cytokine produced by monocytes, macrophages, neutrophils, NK cells, and CD4+ T cells. TNF alpha exhibits cytolytic activity against various tumor cells and plays a key role in immune regulation. It exists as dimers and trimers and also as a 26 kDa membrane-bound form. The MAb11 antibody is a neutralizing antibody.

Applications Reported: MAb11 has been reported for use in intracellular flow cytometric analysis.
Applications Tested: Pre-titrated and tested by intracellular staining and flow cytometric analysis of stimulated human peripheral blood cells. Recommended usage: 5 µL (0.5 µg) per test (final volume 100 µL). Cell number may vary from 10⁵ to 10⁸ cells/test.

Target Information

TNF alpha is a multifunctional proinflammatory cytokine of the TNF superfamily. Secreted mainly by macrophages, it binds to TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR receptors. It regulates immune responses, cell proliferation, differentiation, apoptosis, lipid metabolism, and coagulation. TNF alpha forms multimers and shows a single 17 kDa band under non-reducing SDS-PAGE conditions. Knockout studies indicate neuroprotective functions and tumor necrosis activity. It also plays roles in immune responses to infections and tumor cytolysis, often synergistic with interferon-gamma. TNF alpha is implicated in autoimmune diseases, insulin resistance, and cancer.
