Description:
The MAb11 antibody reacts with human tumor necrosis factor-alpha (TNF alpha), a 17 kDa cytokine produced by monocytes, macrophages, neutrophils, NK cells, and CD4+ T cells. TNF alpha has cytolytic activity against various tumor cells and plays an important role in immune regulation. It forms dimers and trimers and also exists as a 26 kDa membrane-bound form. The MAb11 antibody is a neutralizing antibody.

Applications Reported:
MAb11 has been reported for use in intracellular flow cytometric analysis.

Applications Tested:
Tested by intracellular staining and flow cytometric analysis of stimulated human peripheral blood cells. Recommended ≤1 µg per test (100 µL final volume). Cell number may range from 10⁵ to 10⁸ cells/test. Antibody titration is recommended for optimal performance.

Target Information

TNF alpha is a multifunctional proinflammatory cytokine belonging to the TNF superfamily. It is mainly secreted by macrophages and binds to receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. TNF alpha regulates immune cell activity, proliferation, differentiation, apoptosis, lipid metabolism, and coagulation. It exists as multimers (dimers, trimers, pentamers) and appears as a 17 kDa band under non-reducing SDS-PAGE conditions. Knockout studies in mice suggest neuroprotective roles and tumor necrosis induction. TNF alpha is also involved in immune responses to infections and in the necrosis of certain tumors. Dysregulation is linked to autoimmune diseases, insulin resistance, and cancer.
