Target Information

Ribonucleases (RNAses) H are enzymes that hydrolyze the RNA strands of RNA/DNA hybrids. Their major role is to remove the RNA strand from hybrids formed during DNA replication and repair. RNAse H2 is composed of three subunits, all required for enzymatic activity. Mutations in RNAse H2A or its associated subunits have been linked to Aicardi-Goutieres syndrome (AGS), a neurological disorder with symptoms similar to viral brain infections and elevated IFN-alpha levels in cerebrospinal fluid. Similar pathological features are observed with TREX1 mutations, suggesting that RNAse H2 and TREX1 share roles in preventing accumulation of intracellular nucleic acids and activation of the innate immune system.
