Target Information

DNA methylation, or the addition of methyl groups to cytosine bases in CpG dinucleotides, is essential for proper development and regulates gene expression. Methylation and demethylation are enzymatic processes; the demethylation enzyme is a mammalian protein with demethylase activity associated with a methyl-CpG-binding domain (MBD).
MeCP2 and MBD1 repress transcription by binding to methylated DNA, while MBD2 and MBD4 colocalize with heavily methylated satellite DNA, mediating methylation signal functions. MBD3 does not bind methylated DNA in vivo or in vitro.
MBD1–MBD4 are expressed in somatic tissues; MBD1 and MBD2 expression is reduced or absent in embryonic stem cells. MBD4 shares homology with bacterial DNA N-glycosylases/lyases and can be mutated in microsatellite unstable tumors.
