Target Information

DNA methylation, or the addition of methyl groups to cytosine bases in CpG dinucleotides, is essential for proper development and regulation of gene expression. The methylation pattern involves enzymatic processes of methylation and demethylation. The demethylation enzyme, a mammalian protein with demethylase activity associated with a methyl-CpG-binding domain (MBD), can revert methylated cytosines to cytosines by cleaving the methyl group as methanol.

MeCP2 and MBD1 (PCM1) repress transcription by binding specifically to methylated DNA. MBD2 and MBD4 (MED1) colocalize with heavily methylated satellite DNA and mediate biological functions of the methylation signal. MBD3 does not bind methylated DNA in vivo or in vitro. MBD1, MBD2, MBD3, and MBD4 are expressed in somatic tissues, but MBD1 and MBD2 expression is reduced or absent in embryonic stem cells deficient in MeCP1 activity. MBD4 shares homology with bacterial base excision repair DNA N-glycosylases/lyases and may be mutated in microsatellite unstable tumors.
