Product Specific Information

This NLA14 monoclonal antibody recognizes mouse VSIG4 (V-Set and Immunoglobulin domain containing 4), also known as CRIg or Z39Ig.
VSIG4 is a type I transmembrane glycoprotein related to the B7 family of immune regulatory proteins, containing one complete V-type Ig domain and one truncated C-type Ig domain.
It is expressed on tissue-resident and tumor-infiltrating macrophages and binds complement components C3b and iC3b, inhibiting the alternative complement pathway and promoting phagocytosis.
VSIG4 also suppresses T cell activation, proliferation, and IL-2 production, contributing to immune tolerance and inflammation control.
Expression on tumor-infiltrating macrophages suggests a role in immune evasion.

• Species specificity: Recognizes mouse VSIG4; no cross-reactivity with rat or human VSIG4
• Fixation compatibility: Works with formaldehyde-fixed and permeabilized cells
• Recommended use: Flow cytometric analysis of mouse peritoneal exudate cells
• Optimal concentration: ≤ 1.0 µg/test (100 µL final volume; 10⁵–10⁸ cells/test)
• Excitation/Emission: 405 nm / 600 nm
• Laser: Violet laser

Super Bright 600 is a tandem dye excited by the violet laser line (405 nm) and emits at 600 nm.
Use a 610/20 bandpass filter for detection. Protect samples from light to avoid photo-induced oxidation.

When using multiple Super Bright conjugates, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize polymer interactions.

Samples can be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-57) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.
