Target Information
EIF2AK2 (PKR) is one of four kinases that specifically phosphorylate Ser51 of translation initiation factor eIF2-alpha in response to various environmental stresses. EIF2AK2 plays a key role in antiviral defense by direct activation through double-stranded RNA produced during viral infection. PKR is critical in interferon-induced antiviral responses, virus-induced apoptosis, and regulation of cell growth and differentiation.
PKR induces apoptosis via Fas upregulation and mediates the FADD/Caspase-8 signaling pathway. Upon viral infection, it acts as a dual-function protein, sequentially activating both survival and cell death pathways through kinase-independent and -dependent mechanisms. PKR also regulates NF-kappaB activation, p53, p38, and PDGF signaling pathways and has been implicated in tumor suppression and malignancy. Additionally, PKR is associated with several neurodegenerative diseases, including Alzheimer’s, Huntington’s, and amyotrophic lateral sclerosis (ALS).
