The POLR1D gene encodes a component of RNA polymerase I and III complexes, which are responsible for synthesizing ribosomal RNA precursors and small RNAs. Mutations in this gene are associated with Treacher Collins syndrome (TCS), a craniofacial developmental disorder. Alternative splicing results in multiple transcript variants.
