Target Information

Estrogen Receptors (ER) are members of the steroid/thyroid hormone receptor superfamily of nuclear receptors. The estrogen receptor is a ligand-activated transcription factor that, when bound to estrogen, undergoes conformational change enabling dimerization and binding to estrogen response elements (ERE) in DNA. This interaction allows ER to regulate gene transcription positively or negatively through coactivator or corepressor recruitment.

Two main forms exist: ERα (ESR1) and ERβ (ESR2), encoded by separate genes. Alternative RNA splicing generates at least four ERα isoforms (66 kDa, 53 kDa, 47 kDa, 35 kDa). Estrogen receptors are widely expressed and are crucial for sexual development, reproductive function, and bone health. They are implicated in diseases such as breast cancer, endometrial cancer, and osteoporosis.
