Target Information
Estrogen Receptor beta (ER beta, NR3A2) protein is approximately 55 kDa, though multiple isoforms ranging from 36–59 kDa have been described. Human ER beta was initially cloned and characterized from testis. Estrogen Receptors (ER) are members of the steroid/thyroid hormone receptor superfamily of nuclear receptors.
The estrogen receptor is a ligand-activated transcription factor that, when bound to estrogen, undergoes a conformational change allowing dimerization and binding to estrogen response elements (ERE) in DNA. This binding can regulate gene transcription through coactivator or corepressor recruitment.
Two forms of estrogen receptor exist — alpha (ESR1) and beta (ESR2). ER beta binds estrogens with similar affinity to ER alpha, and some isoforms can inhibit ER alpha activity in reproductive tissues. ER beta is widely expressed across tissues and may act as a tumor suppressor, implicated in conditions such as prostate cancer, autism spectrum disorder, medullary thyroid carcinoma, De Quervain disease, and cardiovascular disease risk.
