Target Information
Apoptosis plays a major role in normal organism development, tissue homeostasis, and removal of damaged cells. Disruption of this process has been implicated in various diseases such as cancer. Members of the Bcl-2 family are critical regulators of apoptosis, characterized by conserved Bcl-2 homology (BH) domains.
A novel member, Bcl-rambo, was identified as a mitochondrial protein whose overexpression induces apoptosis, which can be blocked by co-expression of inhibitor of apoptosis proteins (IAPs) such as XIAP, cIAP1, and cIAP2.
Bcl-rambo shares homology with anti-apoptotic Bcl-2 members but contains a unique 250 amino acid insertion before its C-terminal membrane anchor. This region alone can efficiently induce apoptosis, suggesting that Bcl-rambo may be involved in distinct pro-apoptotic pathways.
