Target Information

Isoform 1 functions as an extracellular chaperone that prevents aggregation of nonnative proteins and stress-induced aggregation of blood plasma proteins. It inhibits formation of amyloid fibrils by APP, APOC2, B2M, CALCA, CSN3, SNCA, and aggregation-prone LYZ variants (in vitro). Clusterin does not require ATP and maintains partially unfolded proteins for subsequent refolding by other chaperones such as HSPA8/HSC70. It protects cells against apoptosis and complement-mediated cytolysis. Intracellular isoforms interact with ubiquitin and E3 ligase complexes to promote proteasomal degradation of target proteins and modulate NF-κB activity. Nuclear isoforms promote apoptosis, while mitochondrial isoforms suppress BAX-dependent cytochrome c release and inhibit apoptosis. Plays a role in regulation of cell proliferation.
