Target Information
Janus kinase 2 (JAK2) is a protein tyrosine kinase involved in cytokine receptor signaling pathways and is mutated in polycythemia vera.
This kinase contains the catalytic domain (JH1) of JAK2. Activation of JAKs leads to phosphorylation of STAT (signal transducers and activators of transcription) proteins, which dimerize and translocate to the nucleus to regulate gene transcription.
JAK2 is essential for IFN gamma-receptor complex initiation, while JAK1 acts as an amplifier. Mutations in JAK2 are associated with disorders such as Budd-Chiari syndrome, Polycythemia vera, Thrombocythemia 3, myelofibrosis, and Acute myelogenous leukemia.
