Product Specific Information

This 37D12 monoclonal antibody reacts with human IL-37 (also known as IL-1F7 or IL-1H4), a member of the interleukin 1 cytokine family.
IL-37 suppresses innate inflammatory and immune responses and exhibits anti-inflammatory characteristics similar to IL-1R8.
It binds weakly to IL-18Rα and does not recruit IL-18β, thus lacking pro-inflammatory signaling.
IL-37 also binds IL-18BP, enhancing its antagonistic effects, forming a trimeric complex (IL-37–IL-18BP–IL-18Rβ) that blocks IL-18 activity.
Expression has been detected in lung, testis, colon tumors, and various human cell lines.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated and validated using the Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00) and BestProtocols® Protocol A.
Use at 5 µL (0.06 µg) per test in 100 µL final volume.
Cell number: 10⁵–10⁸ cells/test (empirically determined).
Excitation: 488–561 nm; Emission: 578 nm; Laser: Blue, Green, Yellow-Green.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

The IL-37 gene encodes a cytokine of the interleukin 1 family that binds to IL18R1 and IL18BP, forming an inhibitory complex with IL18Rβ to suppress IL-18 activity.
Located on chromosome 2 within the IL-1 family gene cluster, IL-37 has five alternatively spliced transcript variants encoding distinct isoforms.
