Target Information

The human UNG gene encodes both nuclear (UNG2) and mitochondrial (UNG1) forms of uracil-DNA glycosylase. These forms arise through alternative splicing and the use of two differentially regulated promoters (PA and PB). The cDNAs for UNG1 and UNG2 are similar in size (2061 and 2058 bp), and therefore appear as a single species on gel electrophoresis.
Nuclear UNG2 differs from mitochondrial UNG1 by 44 amino acids at the N-terminal sequence, which is not required for catalytic activity.
UNG gene products play a major role in repairing mutagenic U:G mispairs caused by cytosine deamination. For example, UNG2 removes misincorporated dUMP residues. Expression of UNG1 and UNG2 varies by cell and tissue type and is regulated by the cell cycle. Generally, UNG expression and activity are higher in proliferating cells compared to nonproliferating ones.
UNG1 is a 304 amino acid protein, and UNG2 is a 313 amino acid protein.
