VISTA is a negative immune checkpoint regulator that suppresses T-cell activation and is highly expressed on hematopoietic cells and within the tumor microenvironment. Similar to PD-L1 blockade, VISTA blockade may provide an immunotherapeutic strategy for human cancer. Therapeutic antibodies against VISTA activate T-cell signaling and promote immune activation.
GenScript Human VISTA Antibody (VA.F6), mAb, Mouse is produced from a hybridoma resulting from the fusion of SP2/0 myeloma and B-lymphocytes obtained from a mouse immunized with recombinant human VISTA.

Target Information

VISTA (V-domain Ig containing suppressor of T-cell activation), also known as B7-H5 or PD-1H, is a product of the VSIR gene. Although VISTA shares sequence homology with PD-L1, it exhibits a distinct expression pattern. In both mouse and human, VISTA is predominantly expressed on hematopoietic cells, with the highest densities on myeloid cells and lower levels on T cells, while being absent on B cells.
VISTA expressed by antigen-presenting cells directly suppresses proliferation and cytokine production of CD4 and CD8 T cells, making it a promising target for cancer immunotherapy. Reported ligands of VISTA include CD28H, VSIG-3, and VSIG-8.
