Product Specific Information

The 30-F11 monoclonal antibody reacts with all isoforms of mouse CD45 (Leukocyte Common Antigen, LCA). CD45 is expressed by all hematopoietic cells except mature erythrocytes and platelets. The cytoplasmic region has tyrosine phosphatase activity, playing a key role in lymphocyte activation.

Each product includes one vial of NovaFluor conjugate and one vial of CellBlox Plus Blocking Buffer.

Note:

• NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal staining.
  • Add 5 µL per sample before mixing with cells.
  • For single-color controls: 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Note: Internal testing shows non-specific B cell staining in SJL mice, not observed in BALB/c or C57BL/6 strains.

Target Information

CD45 (Leukocyte Common Antigen, LCA) is a receptor-type protein tyrosine phosphatase expressed on all nucleated hematopoietic cells (~10% of lymphocyte surface proteins). It is absent in non-hematopoietic tissues. CD45 regulates lymphocyte development and signaling by modulating Src-family kinases and JAK kinase pathways. Multiple isoforms exist due to alternative splicing, differing in extracellular domains but sharing identical transmembrane and cytoplasmic regions. CD45 plays crucial roles in antigen receptor signaling, cytokine receptor regulation, and integrin-mediated survival signaling.
