Product Specific Information

The 30-F11 monoclonal antibody reacts with all isoforms of mouse CD45 (Leukocyte Common Antigen, LCA). CD45 is expressed by all hematopoietic cells except mature erythrocytes and platelets. Its cytoplasmic domain possesses tyrosine phosphatase activity, crucial for lymphocyte activation.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse bone marrow cells at ≤0.4 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Optimize titration for best results.

Important Notes:

• NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell Stains for viability.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining. Add 5 µL per sample before combining with cells.
• For single-color controls: use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology with engineered fluorescent signatures minimizing spillover and spread.
Excitation: 552 nm; Emission: 690 nm; Laser: 561 nm (Yellow).

Note:
Internal testing shows NovaFluor Yellow 690 may non-specifically stain B cells in SJL mice; not observed in BALB/c or C57BL/6 strains.

Target Information

CD45 (LCA) is a receptor-type protein tyrosine phosphatase expressed in all nucleated hematopoietic cells (~10% of lymphocyte surface proteins). It regulates Src-family kinases, T/B-cell receptor signaling, and cytokine receptor signaling. Isoforms (e.g., CD45RA) result from alternative splicing and differ in extracellular domains. CD45 influences cell survival, apoptosis, and immune response modulation.
