Target Information
Beta-catenin은 세포 간 접착 구조(adherens junction, AJ)의 구성 요소로 처음 확인된 단백질입니다. AJs는 세포 성장과 세포 간 접착을 조절하여 상피 세포층의 형성과 유지에 필수적입니다. Beta-catenin은 E-cadherin의 세포질 도메인과 상호 작용하며, E-cadherin을 α-catenin과 연결시켜 E-cadherin 복합체가 피질 액틴 세포골격에 부착되도록 합니다. 또한, beta-catenin은 adenomatous polyposis coli 단백질 및 미세소관을 포함하는 세포골격 복합체와 결합하고, 여러 신호 경로(tyrosine kinase, phosphatase, Wnt/Wingless 등)와 상호 작용합니다. 이 상호 작용은 형태발생을 조절할 수 있습니다.
Beta-catenin은 모낭 세포, 기저 및 주변 기질 세포, 외부 및 내부 뿌리초 세포 등 여러 세포 유형에서 발현됩니다. 또한, pilomatrixoma, medulloblastoma, 대장암, 난소암 등 다양한 종양 발생과 관련이 있습니다. 핵 내에서 beta-catenin은 Lef/Tcf 전사 인자와 함께 작용하여 cyclin D와 c-myc 등 세포 증식을 촉진하는 유전자 발현을 유도합니다. 이러한 작용은 유방, 대장, 간, 폐, 위, 전립선, 피부 등 고형 종양의 발생과 관련된 분자적 기반을 제공합니다.
