Target Information

CD34 is a highly glycosylated monomeric surface protein (111–115 kDa) expressed on various stem cell populations. It serves as a stem cell marker with reversible expression on human hematopoietic stem cells. CD34 may function as a surface receptor involved in receptor-mediated endocytosis and regulation of adhesion, differentiation, and proliferation of hematopoietic stem and progenitor cells.

Its expression likely represents a specific developmental state with distinct adhesion properties. CD34 may act as an adhesion molecule mediating attachment of stem cells to bone marrow extracellular matrix or stromal cells, and as a scaffold for lineage-specific glycans enabling binding to lectins on stromal cells. It also plays a role in presenting carbohydrate ligands to selectins. The intracellular domain is phosphorylated by activated protein kinase C, suggesting a role in signal transduction.

Diseases associated with CD34 dysfunction include dermatofibrosarcoma and neurofibroma.
